The Cover

Immunofluorescent localization of the copper efflux transporter ATP7A (red) in ovarian carcinoma cells. In untreated cells ATP7A is found in the trans-Golgi in the perinuclear region. Exposure to 500 IIMcopper for 1 hour caused ATP7A to redistribute away from the trans-Golgi toward the cell surface (top panel). In contrast, exposure to an equitoxic concentration (50 IIM) of carboplatin for 1 hour had no effect on the distribution of ATP7A, which remained in the perinuclear region (bottom panel). Thus, despite the fact that ATP7A mediates both copper and carboplatin resistance, these compounds have very different effects on the trafficking of ATP7A.
